Background:

HRP-conjugated goat anti-mouse IgG secondary antibody is affinity-purified and is suitable to detect primary antibodies used in WB, IHC and ELISA. First, a primary antibody of mouse origin is bound to a target antigen. After a brief wash, HRP-conjugated goat anti-mouse IgG secondary antibody is applied to form a complex with the primary antibody. After another brief wash, HRP substrates such as luminol or 3,3'-diaminobenzidine (DAB) are added to generate detectable chemiluminescence or insoluble colors.

Antibody Validation

HEK 293 cell lysates were loaded as indicated and separated by SDS-PAGE. Anti-ACTB (β-actin) mouse monoclonal primary antibody (Cat#1170, 1:5,000) and HRP-conjugated goat anti-mouse secondary (Cat#101, 1:20,000) were applied. To detect the signal, the membrane was incubated with PiQ™ ECL (Cat#636), and the image was captured with a ChemiDoc XRS+digital imager.
